守護「蒙娜麗莎」大作戰即將開戰在鳥不生蛋的荒島上過著日復一日生活的流浪貓文森，原以為會這樣平淡無奇地過著他的喵生，沒想到一場洪水之災，不僅讓他與老鼠莫瑞斯成為莫逆之交，也讓兩人意外被運送到擁有超多珍貴館藏的艾米塔吉博物館。
文森在那裡遇到了視老鼠為敵的冬宮菁英貓小隊，他們多年來為了保護博物館的館藏不受老鼠侵害不遺餘力，而一直以來都獨自生活在荒島上的文森，渴望擁有許多貓朋友，但又不想失去救了他一命的死黨莫瑞斯，於是雖然他似乎加入了捕捉老鼠的貓小隊，但暗自卻一直掩護莫瑞斯，不讓他被捕獲。
今晚，世界上最偉大的名畫「蒙娜麗莎」即將被送到博物館，莫瑞斯心癢難耐，這是他一生最夢幻的時刻，只要啃一口「蒙娜麗莎」，他就可以在他的家族寫下歷史。
而正直善良的文森和他的菁英貓小隊夥伴們似乎也做好了守護「蒙娜麗莎」大作戰的準備，卻沒想到一夜之間「蒙娜麗莎」不翼而飛…。
文森能否靠智慧找出名畫的下落，維護博物館的聲譽，並贏得博物館內最美埃及豔后貓的芳心呢？
【關於電影】
一群被飼養在俄羅斯聖彼得堡冬宮博物館（又名艾米塔吉博物館）的貓咪們，是用來驅趕老鼠以保護博物館藏品不被破壞的貓守衛，相傳第一隻貓是由彼得一世親自帶到冬宮，後來其女兒伊麗莎白·彼得羅芙娜頒布了“關於將貓驅逐出宮廷的法令”，但鼠疫越發嚴重才又招募強壯的冬宮貓，之後的葉卡捷琳娜二世授予他們“美術館衛士”的地位，准許他們繼續守護藝術品們。
這些冬宮貓多年來成為博物館的一大特色，吸引了不少愛貓人士慕名而來。
巔峰時期博物館內有高達300隻冬宮貓，現為50隻左右，平時這些貓居住在博物館的地下室，他們有專人照料、以及專屬的貓食堂及貓宿舍，夏天時會出來在堤岸和廣場上走動，在過去他們甚至可以在博物館內的畫廊自由行動。
自1998年起，冬宮博物館將4月6日訂為「貓日」，定期在每年的3月底和4月初間舉辦和貓有關的活動。
2015年，博物館架設了專屬網站，開放給民眾領養冬宮貓，可以將他們帶回家。
